[Chemiluminescence assay for human natural killer cell activity].

چکیده
Recently it has been shown that the natural killer (NK)-target cell interaction results in an immediate burst of chemiluminescence. In this paper we investigated to apply chemilumines cence assay for the evaluation of human NK cell activities. Degrees of chemiluminescence were dependent on the concentration of lymphocytes or tar. get cells, but optimal concentrations of lymphocytes and target cell were 2×106/ml, 3× 106/ml respectively. The responder cell that generates chemiluminescence after target-effecfor interaction is most likely a lymphocyte for a variety of reasons. First, monocyte-depleted fraction containedmore than 95% of lymphocytes. Removal of NK cells with monoclonal anti HNK-1 antibody and complement abolished the chemiluminescence response. Lymphocytes or target cells (K562) generated very little chemiluminescence response. Furthermore, cell to interaction between allogeneic or autologus lymphocytes did not generate any chemi luminescence. A survery of 10 normal peripheral lymphocytes revealed a well correlation (P<0.01) of NK cell activities between chemiluminescence and 51 Cr release assays. These results propose that chemiluminescence assay would be another assay system for human NK cell activities.

Chemiluminescence response of human natural killer cells. I. The relationship between target cell binding, chemiluminescence, and cytolysis
The binding of tumor cells or fetal fibroblasts to human natural killer (NK) cells led to a rapid chemiluminescence response within seconds of target-effector interaction. The degree of chemiluminescence was dependent on the concentration of NK-enriched lymphocytes or target cells, and plasma membrane vesicles from K562 also induced a chemiluminescence response. Mild glutaraldehyde treatment of...
متن کاملIncreased Natural Killer Cells Activity in Schizophrenic Patients
Background: Schizophrenia has been associated with altered immunity. Different studies regarding natural killer cell activity (NKA) in schizophrenic patients have shown inconsistent results. Objectives: To evaluate NK cell activity in schizophrenic patients in comparison with healthy control individuals. Methods: 30 medication-free schizophrenic patients and 41 healthy sex, age and smoking st...
متن کاملThe Influence of Perforin Expression on the Sensitivity of LAK/NK Killing Against Various Tumor Target Cells
Background: Perforin is known to be important in cytolytic activity mediated by natural killer (NK) cells. Objective: To study the relationship between the efficiency of NK and lymphokine-activated killer (LAK) cells activity, and the expression of perforin and HLA class I molecules. Methods: LAK cells were generated by in vitro culturing of human peripheral blood lymphocytes (PBLs) in the ...
متن کاملChemiluminescence Response of Human Natural Killer Cells
Granulocytes and monocytes are known to phagocytose and kill numerous varieties of bacteria and parasites. This function is intimately associated with an early event called the "respiratory burst," which includes the generation of 02and H202 (1). These and other highly reactive molecular species can easily be detected in the presence of luminol or other readily oxidized substrates by the emissi...
متن کاملSUSCEPTIBILITY OF HUMAN WM MELANOMA CELL LINES TO NK AND LAK CYTOTOXICITY AND THEIR RELEVANCE TO THE LEVEL OF MHC CLASS I AND ICAM-l ANTIGEN EXPRESSION
The effect of natural killer (NK) cells and lymphokine activated killer ( LAK) cells was studied on a group of human melanoma cell lines. Peripheral blood from healthy volunteers was utilized as a fresh source of natural killer cells and rhI L-2 for producing LAK cells. The cytotoxicity of effector cells was quantified using a 4 hour SI determining the density of antigen expression on tumor...
